چکیده

The availability of dense panels of common single-nucleotide polymorphisms and sequence variants has facilitated the study of statistical features of the genetic architecture of complex traits and diseases via whole-genome regressions (WGRs). At the onset, traits were analyzed trait by trait, but recently, WGRs have been extended for analysis of several traits jointly. The expectation is that such an approach would offer insight into mechanisms that cause trait associations, such as pleiotropy. We demonstrate that correlation parameters inferred using markers can give a distorted picture of the genetic correlation between traits. In the absence of knowledge of linkage disequilibrium relationships between quantitative or disease trait loci and markers, speculating about genetic correlation and its causes (e.g., pleiotropy) using genomic data is conjectural.

The many faces of pleiotropy.

Pleiotropy is the well-established phenomenon of a single gene affecting multiple traits. It has long played a central role in theoretical, experimental, and clinical research in genetics, development, molecular biology, evolution, and medicine. In recent years, genomic techniques have brought data to bear on fundamental questions about the nature and extent of pleiotropy. However, these effort...

Toward a molecular understanding of pleiotropy.

Pleiotropy refers to the observation of a single gene influencing multiple phenotypic traits. Although pleiotropy is a common phenomenon with broad implications, its molecular basis is unclear. Using functional genomic data of the yeast Saccharomyces cerevisiae, here we show that, compared with genes of low pleiotropy, highly pleiotropic genes participate in more biological processes through di...

On the definition and measurement of pleiotropy.

Pleiotropy, a 103-year-old concept [1], refers to the phenomenon of one mutation affecting multiple traits. Compared with the fundamental importance of pleiotropy in genetics, development, evolution, and medicine, our empirical knowledge of the prevalence and mechanism of pleiotropy is scarce [2]. Given this context, Paaby and Rockman’s recent Opinion article on pleiotropy and its empirical est...

Allelic diversity and association analysis for grain quality traits in exotic rice genotypes

The present research aims to study the association and allelic diversity of linked microsatellite markers to grain quality QTLs of 84 exotic rice genotypes. To this end, 9 microsatellite markers (RM540, RM539, RM587, RM527, RM216, RM467, RM3188, RM246, RM5461) were used in which a total of 61 alleles were identified with a mean of 6 alleles per locus. The polymorphism information content (PIC) ...

Molecular markers as an indicator in the malignant potential of oral lichen planus: A systematic review

BACKGROUND AND AIM: Oral lichen planus (OLP) is a chronic, autoimmune, inflammatory disease. The progression of OLP to oral squamous cell carcinoma (OSCC) has generated a longstanding controversy about the details of this malignant transformation potential. This study aimed to do a systematic review on the molecular markers related to the malignant transformation of OLP.METHODS: We searched the...
